Sanger sequencing workflow
The steps for SeqStudio Sanger sequencing are:
DNA Extraction using Chelex
PCR amplification
Gel electrophoresis
PCR cleanup -column based
(Could also use Enzymatic kit or gel
purification)
Cycle sequencing reaction (Big Dye Terminator 3.1 diluted with BDT sequencing buffer 1.1)
Include a sequencing control in cycle sequencing
BDX cleanup - Big Dye Xterminator bead cleanup
Setup capillary electrophoresis run for BDX based on expected amplicon size
Start the run
Monitor the run
