💡 Application of two-photon Bessel light sheet microscopy to map dopaminergic neurons in adult rat brain.
🧠 The brain hemisphere of an adult rat was cleared using iDISCO tissue clearing protocol. An antibody against the Tyrosine Hydroxylase (TH) was used in combination with Alexa Fluor® 594 to reveal the dopaminergic neurons. The sample was generously donated by Dr. Martin Levesque, and Dr. Christian Ethier, of the CERVO research center, Université Laval.
🔬 Dopaminergic neurons were imaged with an axial resolution of 2 µm using a horizontal SPIM configuration combined with two-photon microscopy and Bessel beam. A wavelength of 840 nm was used for excitation with an exposure time of 50 ms and a 10X (NA 0.6) objective was used for detection.
📏 The image (not representing the entire dopaminergic circuit) measures 5.2 mm x 6.5 mm x 0.5 mm and is composed of 4x5 tiles, assembled with Imaris Stitcher. Each tile is a z-stack of approximately 250 slices acquired with a full field of view (1.3mm x 1.3mm).
