Aging is a nearly universal process that affects all tissues, although it is poorly understood. With aging, our different tissues lose their regenerative capacity, which depends primarily on their stem cells. Skeletal muscle contains a designated population of adult stem cells, called satellite cells, which are generally inactive (quiescent). These cells maintain their regenerative capacity throughout life, but at a very advanced age they fail for reasons that are not well understood. The decisions these cells make during the muscle regeneration process are dictated by both intrinsic and extrinsic (local and/or systemic) mechanisms in a dynamically interactive microenvironment.

This lecture will focus on the role of satellite cells in skeletal muscle regeneration and the dysregulation of their control mechanisms during aging. Strategies to prevent or reverse the loss of function will also be presented, with the aim of advancing towards regenerative therapies.
