Dota 2 | Necro vs Nyx - 554k damage
This is my answer to the Dota 2 above 500k damage in 1 hit or spell puzzle. The official solution is Nyx vs Tusk though. Obviously solutions with stackable something(slark stats, creep health for clinkz, rubic's infinite mana, etc) mechanics excluded.
