Here in Hybridization of probe step we perform colony Hybridizarion technique used for screening of genomic library.
In this step the bacterial colony is transfered to nylon membrane and denatured to expose the naked DNA . Then this membrane is soaked in a solution containing the labelled probe. Then washing is performed to remove nonspecific binding. The nylon sheet is sandwitched on Xray film. The place where Probe is bound is analyzed through autoradiograph and compared with master colony to find the exact segment of genome cloned in that colony.
