You can visit Plzen by car, bus our train directly from Prague. It's a short ride and inexpensive, but I do suggest to pay a little extra and reserve your seats in the train as you may get stuck standing the whole way. I took the train and it was a nice ride.
You can book the brewery tour ahead of time or just purchase tickets on the spot, but check the times of the English tour.
