Example 1 of three-dimensionally reconstructed images of immunostaining of the mouse ear epidermal sheet prepared via the Basic Protocol 1. Epidermal tight junction (ZO-1 staining in green), Langerhans cells (MHC II staining in red), and dendrites of Langerhans cells (claudin-1 staining in blue) visualized by confocal microscopic observation. MHC II of Langerhans cells localizes at perinuclear lesion in steady state. Claudin-1 localizes not only at the dendrites of Langerhans cells but also at tight junctions; however, the blue staining of tight junction is too weak to see in the movie.
