Super-resolution 3D rendered neuron (spinSR10)

This hippocampal mouse neuron (courtesy of Saber Abd-Elkader, PhD student at QBI) was acquired by a 60x magnification oil immersion objective on the new Olympus SpinSR10 system here at QBI. The original image is a multi-tiled montage stack with a voxel resolution of 34 nm in X/Y vs 130 nm in Z, the final image boasting a dimension of 7168 over 4032 pixels across.
The image was acquired using the SoRa disk and the associated 3.2X magnifier, deconvolved in Huygens Pro and rendered in Imaris 9.9.1.
Transfected GFP in green to visualize the arborescence of the neuron. DDHD2 protein stained by IHC and revealed by Alexa 561 secondary antibody in orange.
Both 488nm and 561nm channels are rendered as semi transparent surfaces with emission corresponding colors in Imaris. In addition, the arborescence of the neuron is reconstructed using the filament tracer module displaying the dendrites in white and the spines in blue.
The rendering of the movie was done on one of our Windows workstations optimized by our IT team for graphic acceleration and processing.
