This video present the high-throughput production of a series of 3D reconstructed CA1 pyramidal neurons, stained by Golgi-Cox impregnation method. This is a 3D "digital quantitative" neurons, which includes morphological profile of pyramidal neurons, such as dendritic length, branch orders, spine counts and spine density.

Pyramidal neurons of Golgi-stained sections (150-um thick) were digitized, contoured, followed by spine counts under 100x objective lens (oil immersion) of Zeiss digital upright microscopy using stereology-based image analysis software (Neurolucida) integrated with in-house analysis platform.
