Webinar #22: Aging—The Science of Staying Young
All roads lead to Rome, but there are many roads away from it. Aging is multifactorial—yet Dr. Ed Park argues that stem cell depletion is the central highway to senescence. After 13 years of taking telomerase activators, he explores the science of aging and why exosomes might slow the inevitable decline.

The Three Forces (Trimurti):
Like the Hindu trinity, aging has three aspects: Brahma (creation—stem cells), Vishnu (preservation—telomerase), and Shiva (destruction—senolytic drugs). Most scientists focus on one, missing the whole picture.

Telomere Theory:
Short telomeres correlate with nearly all diseases. Dr. Park's first book, Telomere Time Bombs, and his second, The Telomere Miracle, explore how telomerase activation may slow aging.

Evidence: bristlecone pines live 3,000–6,000 years with long telomeres and high telomerase activity.

Epigenetic Clocks:
Recent research found 13 common methylation sites across all mammals that correlate with aging. But causation remains unclear—progeric children (rapid aging) don't show advanced epigenetic clocks, suggesting it's not causal. Epigenetic copying may be "lossy," accumulating errors like DNA replication.

Stem Cell Depletion:
The real driver of aging: stem cells gradually lose their "stemness"—the ability to be immortal (via telomerase) and asymmetrically divide (one mother, one daughter). When mothers differentiate into two daughters, stemness is lost. Over a lifetime, stem cell niches (retina, hair, pancreas, bone marrow) become depleted and clonally restricted. The 115-year-old Dutch woman had only 2 clonal variants of blood-producing cells instead of millions—she was "discharged."

Dyskeratosis Congenita:
The ultimate aging model. Kids are bald, gray, and prematurely aged. Mutations in any of three genes (TERT, TERC, DYSKERIN) cause the same syndrome, proving telomerase dysfunction drives aging.

Solutions:
1. Stem Cell Banking:
Harvest bone marrow at age 21, cryopreserve it, and thaw it later to replenish depleted stem cells. A $6 billion market projected to double by 2025.
2. Yamanaka Factors:
Four proteins (OCT4, SOX2, KLF4, C-MYC) can reprogram any cell back to pluripotency—proving cells can "lead back to Rome." But p53 (the "watchman") kills cells with DNA errors, preventing runaway de-differentiation.
3. Senolytics (Foxy4dri):
A drug that inhibits FOX04, allowing p53 to kill senescent cells. Early mouse studies show younger coats, better behavior, more hormones, less insulin resistance. But risk: if you kill old cells without young ones to replace them, you're in trouble (the "leaky pail" problem).
4. Exosomes:
Anti-inflammatory, promote de-differentiation, trigger apoptosis, reactivate endocrine axes, reduce autonomic stress. Dr. Park's mom (83) has received 13 treatments and reports better energy, memory, optimism—and wants to rent an RV and drive the lower 48.
Personal Insight:

Dr. Park, now 53, has taken telomerase activators for 13 years. Without them, he feels grumpy and like "day x+1." With them, it feels like "Groundhog Day"—same day, no aging. His mom's transformation from fear of aging to excitement about life suggests exosomes work partly through psychology and hope.

Honest Assessment:
No one knows optimal dosing (some wealthy patients stockpile 70 vials)
IV goes everywhere; nasal is Dr. Park's innovation
Constant immune activation may be counterproductive
Pulses of short regeneration are better than chronic use
Aging is not insurmountable—it's just poorly understood and underfunded

Cost:
$500–$700 per billion exosomes; 5 billion costs ~$3,000 retail (Dr. Park charges less).
