Reports in Nature have stated that pancreatic cancer 5-yr survival rate is a dismal 5%; studies are urgent and eager to identify a more reliable treatment for this typically late-stage detected malignancy. Nearly 90% of all pancreatic malignancies can be attributed to the subtype of pancreatic ductal adenocarcinoma, which ranks 4th overall in cancer-related fatalities. PANC-1 cells were first established from the primary pancreatic ductal tissue of a Caucasian 56-yr old male patient diagnosed with pancreatic epithelioid carcinoma. PANC-1 cells are well characterized and a well-established pancreatic cancer model system with well-developed therapeutic assays. PANC-1 is aptly named after the pancreas and is considered a suitable transfection host. PANC-1 cells are also inhibited in growth by L-asparaginase and have been used in non-endocrine tumorigenicity studies. PANC-1 cells overexpress the HER2/neu oncogene, are ER negative, MUC4 negative, Smad4 positive, and have the type B G6PD phenotype. The HER2 oncogene can be found in 60-70% of pancreatic carcinomas. FGF2 growth factor stimulation can induce differentiation of PANC-1 cells into islet-like clusters that produce hormones. PANC-1 cells have a modal chromosome number of 63 and a doubling time of roughly 52 hours.
