MRC5 Cell Line:
In biomedical research, human diploid cells are frequently used for purposes such as culturing viruses for vaccine production. Diploid cells show a comparatively higher safety and immunogenicity than other tissue cultures. MRC-5 cells are the most widely used human diploid cell line for current human viral vaccine manufacturing. The MRC-5 line was first established in 1966 by J.P. Jacobs and colleagues from pulmonary tissue of an aborted male 14-week-old human fetus. MRC5 cells display a modal chromosome number of 46 and an XY karyotype. Polyploidy occurrs at a rate of 3.6%. MRC-5 cells are adherent in culture and can have 42-46 population doublings before the onset of senescenc. MRC-5 cells are negative for reverse transcriptase, and are capable of supporting a broad range of virus growth including adenoviruses, coxsackie a, herpes simplex virus, cytomegalovirus, echovirus, poliovirus, respiratory syncytial virus, rhinovirus, and varicella-zoster virus. The MRC-5 cell line has been used in studies such as virology-related transfection experiments, assessment of cytotoxicity, and the vaccine development; MRC-5 cells are suitable as a transfection host, in part due to their ability for easy viral infections.
