In this video, I will teach you about Alpha-2 Agonist pharmacology, including their mechanism of action, types, indications, adverse effects, contraindications, special considerations and drug-to-drug interactions.

Alpha-2 agonists bind to alpha-2 receptors on sympathetic pre-synaptic nerve terminals, reducing norepinephrine release and, thus, sympathetic nervous system activity. The most common are clonidine, dexmedetomidine, guanabenz, guanfacine and methyldopa. They are most commonly used to treat hypertension but also treat severe pain, ADHD, and withdrawal symptoms from substances. The most common adverse effects are sedation and xerostomia, but they can also cause bradycardia and hypotension. They should be used cautiously in those older than 65. Blood pressure and heart rate monitoring are important during therapy. If used for hypertension, the dose can be taken at bedtime to avoid daytime sedation. Use cautiously with central nervous system depressant, anti-hypertensive and negative chronotropic medications.
